Implication of Genetic Factors in Neural Tube Defects Etiology

چکیده
Neural tube defects are a group of developmental abnormalities that results from failure of fusion of the caudal neural tube, representing one of the most common group of malformations of human structure. The etiology is thought to be multifactorial in the vast majority of the cases, but other possible causes are heterogeneous and include chromosome abnormalities, single gene disorders and teratogenic exposures. However, the exact cause is not known in most cases and etiology remains rather complex and poorly understood. The aim of this study was to provide evidence for a genetic contribution to the etiology of neural tube defects. Multifactorial defects have a significant genetic component that interacts with specific environmental factors. Our study aimed to identify some of these risk factors, but it also established an association between the occurence of neural tube defects with spontaneous abortions, aswell as with other congenital defects, that may be considered markers of early prenatal damage. Concordance of these defects in families also sustained implication of genetic factors in their etiology.

Classification, clinical features, and genetics of neural tube defects
Neural tube defects (NTDs) constitute a major health burden (0.5-2/1000 pregnancies worldwide), and remain a preventable cause of still birth, neonatal, and infant death, or significant lifelong handicaps. The malformations result from failure of the neural folds to fuse in the midline, and form the neural tube between the third and the fourth week of embryonic development. This review article ...
متن کاملNeural Tube Defects, Its Etiology: Environmental Exposures and Genes, Possible Risk Factors
Aim The etiology behind abnormal development of the neural tube during embryonic life is not well understood. The evidence for associations of environmental cause and genes remains controversial. However recent analysis suggests excessive corresponding risks at environmental exposure levels and involvement of genes. Methods We analyzed the research on developmental defects of neural tube and de...

Neural tube defects (NTDs) are among the most common complex congenital malformations observed in newborns. When the neural tube fails to close completely, severe malformations of the brain and/or spinal cord and subsequent neurologic impairment occurs. It is widely believed that nutritional, environmental and genetic interactions contribute to NTDs. It is well established that low folate level...
متن کاملRisk factors for neural tube defects: a study at university-affiliated hospitals in Tehran.
BACKGROUND Neural tube defects have a complex and imperfectly understood etiology involving both genetic and environmental factors. In this case-control study, we tried to determine the association of known risk factors for neural tube defects in patients referred to university-affiliated hospitals with neurosurgical services in the city of Tehran. METHODS Risk factors were assessed in-person...
متن کاملGenetic evidence in planar cell polarity signaling pathway in human neural tube defects.
Neural tube defects (NTDs) are a group of birth anomalies having a profound physical, emotional, and financial effects on families and communities. Their etiology is complex, involving environmental and genetic factors that interact to modulate the incidence and severity of the developing phenotype. The planar cell polarity (PCP) pathway controls the process of convergent extension (CE) during ...
